Sinopse: Michael Scofield (Wentworth Miller, "Joan of Arcadia") é um homem desesperado numa situação desesperadora. Seu irmão, Lincoln Burrows (Dominic Purcell, "John Doe"), está no corredor da morte e será executado em alguns meses, após ser condenado por um assassinato que Lincoln não cometeu. Sem outras opções e com o tempo diminuindo, Michael assalta um banco para que ele seja preso e levado para a penitenciária estadual Fox River, o mesmo local onde seu irmão está cumprindo pena. Uma vez lá dentro, Michael — um engenheiro civil com as plantas da prisão — começa a executar um elaborado plano para libertar Lincoln e provar a inocência dele. Com a ajuda de seu companheiro de cela, Sucre (Amaury Nolasco), Michael começa a se aliar a um grupo de diferentes prisioneiros, incluindo o ex-chefão da máfia John Abruzzi (Peter Stormare), "(Minority Report)" e Charles Westmoreland (Muse Watson, "(Eu Sei O Que Vocês Fizeram No Verão Passado)", um homem que muitos acreditam ser o famoso ladrão D.B. Cooper. No lado de fora, Michael tem apenas uma aliada, sua advogada e amiga de longa data Veronica Donovan (Robin Tunney), "(Fim dos Dias)" — que também é a ex-namorada de Lincoln. Entre os personagens secundários também estão a Dra. Sara Tancredi (Sarah Wayne Callies, "(Tarzan)", que Michael acaba visitando por diversas vezes na enfermaria do presídio; o policial Bellick (Wade Williams), "(Colateral"), que está determinado a ficar de olho em seu novo prisioneiro; e o poderoso Warden Pope (Stacy Keach, "Titus"), que finge ter um relacionamento próximo, quase uma relação entre pai e filho, com seu novo condenado. "Prison Break" tem semelhanças com diversos filmes, combinando a esperança de "Um Sonho de Liberdade", a camaradagem de "Golpe Baixo" e a tensão de "Fugindo do Inferno".
